The 68-year-old's return has been the subject of considerable speculation in recent days.
Considered as one of the "big beasts" of the Conservative Party, Mr Clarke will take on the business and enterprise portfolio in a shadow cabinet reshuffle.
He said he was inheriting the post of current shadow business secretary Alan Duncan to help Britain tackle "the gravest economic crisis I have known in my lifetime".
"It is going to be a historically important election, and I don't want to sit on the sidelines - I want to be out on the pitch fighting for the change Britain needs," he said in a statement.
